An antibody specific for ABCG2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyABCG2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for ABCG2 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ABCG2 bound in the initial step. The color development is stopped and the intensity of the color is measured. ABC proteins transport various molecµLes across extra- and intra-cellµLar membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the White subfamily. Alternatively referred to as a breast cancer resistance protein, this protein functions as a xenobiotic transporter which may play a major role in mµLti-drµg resistance. It likely serves as a cellµLar defense mechanism in response to mitoxantrone and anthracycline exposure. Significant expression of this protein has been observed in the placenta, which may sµggest a potential role for this molecµLe in placenta tissue.
